Measuring the Binding Constants with Use of SPR
Biacore T100 (GE Healthcare) is a biomolecular interaction analyzer that uses Surface Plasmon Resonance (SPR) electrooptical phenomenon to measure the binding constants and on/off rates between binding partners. It is a versatile system for high-quality characterization of molecular interactions, from ions to viruses. One platform is applicable for comprehensive characterization of molecular interactions - in terms of kinetics, affinity, specificity, comparability, concentration and thermodynamics.
The ligand is typically immobilized on a surface of a gold chip, and a solution of analyte is passed over the surface. The changes in SPR angle being measured are related to affinity constant and to the ratio of molecular masses of analyte to ligand.
Therefore, Biacore is a powerful instrument that enables label-free kinetic and thermodynamic analysis of monovalent and polyvalent binding.
